PENGARUH PENAMBAHAN SARI DAUN SIRIH SEGAR KE DALAM PENGENCER SITRAT KUNING TELUR TERHADAP KUALITAS SEMEN CAIR BABI PERSILANGAN LANDRACE DAN DUROC

Abstrak

Penelitian ini dilakukan dengan tujuan untuk mengkaji pengaruh penambahan sari daun sirih segar kedalam pengencer sitrat kuning telur terhadap kualitas semen cair babi hasil persilangan landrace dan duroc. Semen berasal dari satu ekor babi pejantan hasil persilangan landrace dan duroc berumur 1,5 tahun dan dalam kondisi sehat. Semen dievaluasi secara makroskopis dan mikroskopis. Metode penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan enam perlakuan dan lima ulangan yang terdiri atas P0= Sitrat Kuning Telur (SKT), P1= SKT+ Sari Daun Sirih (SDS) 0,5%, P2= SKT+ SDS 1%, P3= SKT+ SDS 1,5%, P4= SKT+SDS 2%, P5= SKT+ SDS 2,5%. Semen segar yang digunakan pada penelitian ini memenuhi standar dengan motilitas 81,00% dan abnormalitas 3,20%. Semen yang telah di encerkan kemudian dibagi kedalam tabung eppendof lalu disimpan dalam kotak Styrofoam bersuhu 15º - 20ºC dan diamati setiap dua belas jam sekali. Data yang diperoleh dianalisis menggunakan Analisis of Variance (ANOVA) dan uji lanjut Duncan dengan menggunakan program SPSS 26.0. Hasil penelitian menunjukkan bahwa persentase motilitas pada perlakuan ke-5 jam ke 48 adalah 48.80±5.70 lebih tinggi dari pada P0, P1, P2 P3 dan P4, dan memiliki persentase viabilitas spermzatozoa: 61.10±5.10, abnormalitas spermatozoa: 6.60±0.42 dan daya tahan hidup: 48,00±1,57 jam. Disimpulkan bahwa penambahan sari daun sirih dalam pengencer sitrat kuning telur efektif dalam mempertahankan kualitas semen babi hasil persilangan landrace dan duroc sampai 48 jam penyimpanan, dengan dosis terbaik Sari Daun Sirih (SDS) 2,5%.

This study was conducted to explored the effect of adding fresh betel leaf extract into egg yolk citrate diluent on the quality of liquid semen of crossbred landrace and duroc pigs. Semen came from one male pig of crossbred landrace and duroc aged 1.5 years and in good health. Semen was evaluated macroscopically and microscopically. The research method used a Completely Randomized Design (CRD) with six treatments and five replications consisted of P0 = Egg Yolk Citrate, P1 = EYC + Betel Leaf Extract 0.5%, P2 = EYC + BLE 1%, P3 = EYC + BLE 1.5%, P4 = EYC + BLE 2%, P5 = EYC + BLE 2.5%. Fresh semen used in this study met the standards with 81.00% motility and 3.20% abnormality. The diluted semen was then divided into eppendorf tubes and stored in a Styrofoam box at a temperature of 15º - 20ºC and observed every twelve hours. The data obtained were analyzed using Analysis of Variance (ANOVA) and Duncan's further test using the SPSS 26.0 program. The results showed that the percentage of motility in the 5th hour of treatment 48 was 48.80 ± 5.70 higher than P0, P1, P2 P3 and P4, and had a percentage of sperm viability: 61.10 ± 5.10, spermatozoa abnormalities: 6.60 ± 0.42 and survival: 48.00 ± 1.57 hours. It was concluded that the addition of betel leaf extract in egg yolk citrate diluent was effective in maintaining the quality of semen from crossbred landrace and duroc pigs for up to 48 hours of storage, with the best dose of Betel Leaf Extract (BLE) of 2.5%.
